INTESTINAL ABSORPTION AND DIMINUTION IN THE BLOODSTREAM OF GLUTATHIONE IN RABBIT

Intestinal absorption and metabolism of GSH was investigated in the rabbit. The new technique of intravenous injection in the portal vein was employed, GSH was directly detected from the portal vein several times during thet time course after duodenal instillation, and the portal circulation was never hindered during the experiment. GSH was determined by the enzymatic cycling method. GSH absorbed by the intestinal tract was detected in portal plasma and indicated 13.23±1.92 μg/ml at five min after duodenal instillation ; it could be detected up to 40 min after administration. GSH accumulating in the portal plasma was about 3% of the administered GSH. Intestinal absorption of GSH was confirmed. Diminution of GSH in blood passed through the portal vein system was examined. For detection of GSH in carotid artery and jugular vein, more than 2.34 mg and 4.67 mg of GSH, containing 40 μg/ml and 80 μg/ml in portal blood, had to be infused into the portal vein, respectively. These results showed that orally administered GSH was absorbed by the intestinal wall and reached the liver, but it could not be found in the peripheral bloodstream as free GSH.
