Effects of immunoglobulin therapy in experimental allergic neuritis (EAN) of the Lewis rat induced by P2 myelin protein

Background : Experimental allergic neuritis (EAN) is an animal model for the Guillain-Barre syndrome (GBS). Intravenous immunoglobulin (IVIg), a potent modulator of pro-inflammatory assaults, is effective for GBS, but its mechanism is not well understood. Materials and Methods : We tested whether immunoglobulin (Ig) treatment accelerates recovery of disease severity in EAN. EAN was induced in Lewis rats using a bovine P2 myelin protein. Results : EAN rats, with Ig treatment begun just after the onset of clinical symptoms, reduced the clinical, electrophysiological and pathological severity of the disease. In Lewis rats EAN, secondary axonal degeneration due to primary demyelination, was a major pathological feature. Secondary axonal degeneration in EAN Lewis rats with Ig treatment was less severe than in non-treated EAN rats. Conclusions : Ig treatment had accelerated recovery of retarded peripheral nerve conduction, and also reduced the amount of axonal degeneration and demyelination. Ig appears to suppress the active stage in the acute stage of EAN.
