Distribution of transient receptor potential vanilloid 1(TRPV1)in the trigeminovascular system of the rat

Objective : To elucidate the involvement of transient receptor potential vanilloid 1 (TRPV1) in the pathophysiology of migraine, we investigated the distribution of this receptor in the trigeminovascular system of the rat. Methods : A total of 12 Sprague-Dawley rats, weighing 350-450g, were perfused with Zamboni's fixative. The trigeminal ganglia (TG), dorsal root ganglia (DRG), medulla and cervical segments of spinal cord, the circle of Willis with its major vascular branches were removed. The specimens were sectioned at a thickness of 10μm, and the pial arteries were processed as the whole mount preparation. TRPV1 immunoreactivity was detected by goat anti-rat VR1 receptor antibody as 1 : 1000. Results : TRPV1 was observed in the neurons of TG, DRG and the nerve fibers on the dura mater and around the pial arteries and in the trigeminal spinal tract. Conclusions : From our observation, TRPV1 was observed in the trigeminovascular system, and this receptor is likely to be involved in the migraine pathogenesis.
