Determination of Amphotericin B in Human Serum by a High-Performance Liquid Chromatographic Method(Analytical,Chemical)

A highly sensitive method was developed for the determination of serum amphotericin B (AMB), an antifungal antibiotic having a polyene structure, by high-performance liquid chromatography (HPLC) on a column packed with reversed-phase μBondapak C_<18>. Elation was performed with 10mM acetate buffer (pH 7.0)-acetonitrile (61:39, v/v) at a flow rate of 1.5ml/min, with detection at 405nm. The determination limit of AMB was 0.0125 μg/ml in serum. The coefficient of variation for the within-day precision was 5.3-7.6% at 0.025-1.000μg/ml serum. Extraction of AMB from serum samples was 89-102%. The decay curves of serum AMB were determined for six diabetic patients with urinary fungal infections. The mean pharmacokinetic parameters, i.e., peak time, peak level, elimination half-life and absorption half-life were 6h, 0.108μg/ml, 15.1 and 3.5 h, respectively. This method makes it possible, for the first time, to monitor serum AMB levels in patients who have received the drug orally.
