Influence of the Nature of Dentin Surfaces on Proliferation and Mineralization Activity of Gingival and Periodontal Ligament Cells

本研究の目的は,象牙質の表面性状が歯肉細胞と歯根膜細胞の増殖および石灰化能にどのような影響を及ぼすかを検討することである。牛歯から象牙質ブロックを作製し, 3群に分けて次の表面処理を行った。1) Rp群:ハンドスケーラーでルートプレーニング, 2) CA群:pH1のクエン酸で3分間処理, 3) Na群: CA群をさらに10%NaOClで3分間処理。さらにhydroxyapatiteブロックを作製しHA群とし,計4群とした。各ブロック上にヒト歯肉細胞(HGC),歯根膜細胞(HPLC)を播種,培養し, 1, 3, 5, 7, 10, 14日目にアルカリ性フォスファターゼ(ALP)活性と蛋白量を計測した。次にHGCとHPLCを培養し, confluentになった時点で上記4群のブロックを静置, 100nM dexamethasone, 10mM β-glycerophosphateを添加したαMEMでさらに培養し, 1, 2, 3週後にvon-kossa染色して,ブロック辺縁に形成された石灰化結節の面積を測定した。その結果,蛋白量の増加はHGCとHPLCを比較すると4群ともHGCが早く, 4群間で比較すると, HPLCではCA群が早く, HGCではNa群が遅かった。ALP活性はHGCにもわずかに認められたが, HPLCの方が4群とも明らかに高く, 4群間で比較するとHPLCではCA群が最も高くかった。石灰化結節の面積はHPLCがHGCより大きく, 4群間で比較するとHPLCではCA群が大きく, HGCでは差なかった。以上の結果から,象牙質の表面性状はHPLCの増殖と石灰化能, HGCの増殖に影響を及ぼすことが明かとなった。さらに, HGCは低いながら石灰化能を持ち,石灰化物を形成できる可能性を持つことが示唆された。
