Examination of growth promotion factors for smooth muscle cells of hypertrophied bladders in rats following partial outlet obstruction

It was reported that outlet obstruction increased the bladder weight in comparison with the controls from one week after the obstruction. This increase continued until 20 weeks after the operation ; however, there was no increase from 30 weeks post operation. Pathologically, thickening of the wall (mainly the muscle layer) was seen. Previously we reported that soluble fractions dervided from bladder 3 to14 weeks after outlet obstruction facilitated the proliferation of cultured smooth muscle cells in the bladder of a normal rat. In order to identify the growth factors contained in the soluble fractions, we have added various growth factors to cultured normal bladder smooth muscle cells. Only basic-fibroblast growth factor (b-FGF) facilitated the growth of the smooth muscle cells. Further, using RT-PCR method, we examined which growth factors were expressed in hypertrophied bladder, and found the expression of b-FGF, nerve growth factor (NGF) and transforming growth factor-β (TGF-β). As to the reason why only b-FGF promoted growth of the cells in our study, we have to further examine the characteristics of cultured normal bladder smooth muscle cells and their growth environment.
