IDENTIFICATION OF HEPARIN-DEPENDENT CELL ADHESION SITE WITHIN LAMININ α4 CHAIN C-TERMINAL GLOBULAR DOMAIN

Fibroblasts bound to α4LG1-5 and α4LG4 recombinant protein. In heparin dependent manner. Heparin blocked cell adhesion. Inhibition assays showed A4G82;TLFLAHGRLYFN and A4G83;LVFMFNVGHKKL within α4LG4 was active for cell adhesion. This site was previously identified as a heparin binding site by us. (J. Biol. Chem 2000, 275. 29458〜29465, H. Yamaguchi, H. Yamashita, Y. Kitagawa et al.) The synthetic peptide containing A4G90;GAAWKIKGPYIL also showed inhibitory activity. Stable cell lines derived from 293T cells, which express syndecan-2 or syndecan-4 were able to bind to α4LG4 recombinant proteins. These results suggest that cell adhesion to α4LG4 is mediated by cell surface heparan sulfate proteoglycans, syndecans.
