Immuno-Gold Localization of Nitrogenase in Root Nodules of Elaeagnus pungens Thunb

The molybdenum-iron component of nitrogenase (MO-Fe component) was purified from soy-bean nodule bacteroids and antibody was raised against it in rabbits. Antibody raised against the 53 kDa polypeptide which was the major protein in the Mo-Fe component fraction of soybean nitrogenase was confirmed to be specific to the nitrogenase by immunodiffusion and immuno-titration. The nitrogenase from root nodules of Elaeagnus pungens cross-reacted with the antibody and appeared from the results of the immunodiffusion to be partially identical to soybean nitrogenase. Using the antibody, we examined intracellular localization of nitrogenase in root nodules of Elaeagnus pungens, in which Frankia is present as a symbiont, by immuno-gold labelling. Thin sections of nodules of Elaeagnus pungens were first treated with anti-nitrogenase specific antibody and then with colloidal gold-protein A as a marker. The gold particles were observed to be concentrated in the vesicles of the endophyte Frankia. This provides strong support for the existence of nitrogenase in the vesicles. Furthermore, our results suggested that nitrogenase localizes in the hyphae of the endophyte Frankia in Elaeagnus pungens nodules.
