The reaction of a new antitumor platinum complex, (-)-(R)-2-aminomethylpyrrolidine(1,1-cyclobutanedicarboxylato)platinum(II) (1), with guanosine (Guo) in an aqueous solution was kinetically compared with the reactions of carboplatin (2) and cisplatin (3) with Guo using high performance liquid chromatography (HPLC) analyses and proton nuclear magnetic resonance (^1H-NMR).Reactions of 1 and 2 with Guo proceeded according to second order kinetics; the 1,1-cyclobutanedicarboxylate (CBDCA) group of 1 and 2 was directly substituted with two N-7 of Guo.The reaction of 3 followed a two-step course through the intermediate, cis-Pt(NH_3)_2Cl(Guo).The reaction rates of the three platinum complexes with Guo were related to 3>>1>2. This result indicated that the CBDCA ligand (chelate ligand, bidentate ligand) coordinated to platinum more strongly than the Cl ligand (monodentate ligand), and the trans effect of the 2-aminomethylpyrrolidine ligand was stronger than that of the ammine ligand.
