Changes in density of cytochrome P450 2B1/2 molecules in the membrane of endoplasmic reticulum in hepatocytes of phenobarbital-treated rats

To examine how the induction of CYP2B1/2 molecules in the rat liver after phenobarbital (PB) treatment altered the density of the molecules in the membrane of endoplasmic reticulum (ER), we measured CYP2B1/2 content in hepatocyte cytoplasm by image analysis and ER area in the cytoplasm by morphometry, then, CYP2B1/2 density in the ER membrane was estimated. In the animals intraperitoneally injected singly with PB (80 mg/kg body weight), CYP2B 1/2 content increased 7-fold in perivenular (PY) and midzonal (MZ) hepatocytes. ER area increased 1.5-fold in PY hepatocytes but did not increase in MZ hepatocytes. Both the content and ER area in periportal (PP) hapatocytes remained unchanged. In the animals repetitively injected with PB (80 mg/kg) once a day for 3 days, the content increased 15-fold in PV, 18-fold in MZ and 12-fold in PP hepatocytes. ER area increased 2-fold in PV, MZ and PP hepatocytes. Density of CYP2B1/2 in the animals singly injected with PB increased 7-fold in PV and MZ hepatocytes but remained unchanged in PP hepatocytes, whereas the density in the animals repetitively injected with PB increased 9-fold in Pv, 10-fold in MZ and 12-fold in PP hepatocytes.
