Biochemical and Electron-Microscopic Evidence for Membrane Injury in Yeast Cells Quickly Frozen with Liquid Nitrogen

We obtained basic information on verifying the mechanisms of injury caused by intracellular ice formation from fast cooling with liquid nitrogen (LN_2) in yeast. Suspensions of the liquid nitrogen-treated yeast cultures containing [^3H]-labeled deoxy-ribonucleic acid were incubated with added DNase in a buffer solution. Extracellular radioactivity levels increased in solutions of the LN_2-treated cells, although the activities for control cells treated at 0℃ were almost unchanged. We considered that the membrane systems of the LN_2-treated cells were damaged and the permeability barrier destroyed, but that this was not the case in 0℃-treated cells. Furthermore we suggested that native DNase worked effectively on its DNA after the LN_2-treatment. These considerations were confirmed by electron microscopy. Thus, While the ultra structure of 0℃-treated cells was not changed, the cell membrane and nuclear membrane of the LN_2-treated cells were seriously damaged.
