Biosynthetic Pathway of 5-Hydroxymethylcytosine by Streptoverticillium rimofaciens : Biosynthetic Studies on Mildiomycin (III)

A newly discovered biochemical pathway for the formation of 5-hydroxymethyl-cytosine (HMC) from cytidine 5'-monophosphate (5'-CMP) was further examined using a cell-free extract of Streptoverticillium rimofaciens, a mildiomycin producer. In this extract, we found an enzyme activity that causes the formation of HMC from 5-hydroxymethylcytidine 5'-monophosphate (HMCMP) by the hydropysis of the N-ribosyl bond of HMCMP. The hydrolysis reaction was specific for HMCMP and its deoxyribonucleotide (HMdCMP), but not for their nucleosides. The microorganism did not degrade CMP and deoxycytidine 5'-monophosphate into cytosine. The enzymes, CMP hydroxymethylase, the HMCMP-cleavage enzyme (HMCMP N-ribotidase), and 5'-nucleotidase were separated from each other by gel filtration using Bio-Gel P-300. The product of CMP hydroxymethylase was confirmed to be HMCMP by HPLC analysis. Thus, the newly discovered biochemical pathway for HMC formation involved two sequential enzyme reactions : the first, HMCMP formation from CMP and the second, hydrolysis of HMCMP into HMC.
