Microbial Production of Mildiomycin Analogues by the Addition of Cytosine Analogues : Biosynthetic Studies on Mildiomycin (I)

Mildiomycin (MIL), produced by Streptoverticillium rimofaciens, is an antibiotic with a 5-hydroxymethylcytosine (HMC) moiety. Among various cytosine analogues, HMC, when added into the fermentation medium, promoted the production of MIL. Contrariwise, cytosine suppressed MIL production; a MIL analogue (MIL-C) having a cytosine instead of the HMC moiety of MIL was produced together with a significant amount of HMC. In the same way, 5-methyl-, 5-bromo-, 5-iodo-, and 5-fluorocytosine gave rise to the MIL analogues, MIL-M, MIL-B, MIL-I, MIL-F, respectively, whereas MIL production was suppressed. These MIL analogues showed strong antifungal activities against the powdery mildew.
