Formation of 5-Hydroxymethylcytosine from Cytidine 5'-Mono-Phosphate by Streptoverticillium rimofaciens : Biosynthetic Studies on Mildiomycin (II)

The biosynthetic pathway of 5-hydroxymethylcytosine (HMC), a molecular constituent of mildiomycin, was examined in a cell-free system. HMC was enzymatically formed in ppotassium phosphate buffer (pH 8) from cytidine 5'-monophosphate (CMP) and formaldehyde or L-serine in the presence of tetrahydrofolate. The reaction product was identical with authentic HMC on high performance liquid chromatography, ultraviolet abosrption, elementary analysis, thin layer chromatography, and ^<13>C-NMR spectroscopy. The ^<13>C-NMR spectra also revealed that the carbon atom of the hydroxymethyl group of HMC was derived from formaldehyde in the reaction mixture. Thus, a new biochemical pathway for the biosynthesis of HMC, involveing a new enzyme, CMP hydroxymethylase, that catalyzes the hydroxymethylation of the 5-position of CMP, was assumed.
