EFFECTIVE EXTRACTION AND RADIOIMMUNOASSAY OF CHORIONIC GONADOTROPIN IN HUMAN URINE

24時間尿中から,徴量存在する絨毛性性腺刺激ホルモン(hCG)を1回の操作によって高収率で抽出濃縮する方法を検討した.方法はピタチェンジ(パームチット)を担体とした小型カラムによる吸着クロマトグラフィーを利用したもので,そのhCG回収率は定量的に90%以上であり,従来から一般的に用いられているカオリン-アセトン法が約50%であるのに比較して大きく改良された.さらに抽出物中の非特異的な尿蛋白質の混入も低くなった.従ってこの新しい抽出法による尿中徴量hCGの部分精製濃縮法と,先に改発されたhCGβサブユニットの特異的カルボキジル末端ペプチドに対する抗体を用いたラジオイムノアッセイを組み合わせることによって,hLHとの交差性を考慮することなく,尿中hCGの鋭敏かつ特異的な検出が可能となった.最近,正常人各組織中のhCG様物質の存在が報告され,重要な問題となっているため,この新しいアッセイ系を臨床応用するに際し当然だがらhCG値の正常範囲を決定する必要がある.正常人について尿中のhCG様免疫活性を,この新しいアッセイ系で測定したところ,1人に最高値1日当り1.1IUが検出されたが,一般に93%の人が1日当り0.5IU以下の値を示した.このことから,1日当り1.1IU以下のhCG値を一応正常範囲とみなしうると考えられる.この新しい尿中hCGの徴量測定法は,絨毛性疾患愚老のより厳密な管理に有用性が高いものと思われる.
