<SHORT REPORTS>Inhibition of Myosin Light-Chain Kinase Affects Myosin Organization of Preimplantation Mouse Embryos

In order to elucidate the possible role of myosin light-chain kinase (MLCK) in preimplantation mouse development, whole-mount, myosin-specific immunofluorescence cytochemistry was performed in preimplantation mouse embryos after treating the embryos with ML-9, a specific inhibitor fo MLCK. Confocal laser scanning microscopy during the late morula stage revealed the periodic staining pattern of myosin organization in the control embryos. In contrast, the embryos treated with 10μM ML-9,revealed loss of this organization coinciding with decompaction of the embryos.The significance of these observations is discussed.
