<SHORT REPORTS>Effects of MI-9, a Specific Inhibitor of Myosin Light-Chain Kinase, on Preimplantation Mouse Development

Involvement of myosion light-chain kinase(MLCK), a Ca^<2+>/-calmodulin-dependent kinase, in preimplantation mouse development was studied using ML-9 which is a specific inhibitor of MLCK. Continuous teratment 2-cell stage embryos with KL-9 at 0,1,5,10 and 15 μM showed that the effect of this inhibitor on cell proliferation is dose dependent. When treated stage-specifically the embryos showed stage-specific responses which were reversible. Although 10 μM ML-9 had no effect on 4-cell stage embryos, but after the 8-cell stage it interfered with the process and maintenansce of compaction. Cell counting at day 4 of culture of these stage-specifically treated embryos showed that the inhibition of MLCK affects cell proliferation after the 8-cell stage. Therelative specificity of the effects of ML-9 on early mouse embryos suggests that MLCK may play an important role in the process of compaction and cavitation.
