Estimation of the Size Distribution of Lipoproteins by Convenient Gradient Gel Electrophoresis

We developed a convenient method of gradient gel electrophoresis (GGE) using commercially available pre-casted gels to estimate the size distribution of lipoproteins. Serum samples were used after pre-staining with Sudan black B. Each GGE pattern had two broad bands of HDL and a sharp band of LDL. By this method, we studied the relationship between increase of small density LDL and disturbances of lipid metabolism. Small density LDL was increased in 4 out of 6 patients with type IIb hyperlipoproteinemia and 10 out of 15 patients with type IV hyperlipoproteinemia, but not in 7 patients with type IIa hyperlipoproteinemia or 18 normal subjects. Small density LDL was also incresed in 3 out of 6 patients with hypolipoproteinemias. Small density LDL may be caused by an imbalance in lipid transfer among lipoproteins involved in hypertriglyceridemia as well as hypolipoproteinemias.
