Hypotensive effects of KB-944 in patients with essential hypertension.

The effects of KB-944, a new long-acting calcium antagonist, on diurnal variation of blood pressure were investigated in 37 inpatients with essential hypertension . The study periods consisted of an observation period and a treatment period. KB-944 30 mg per day was administered once a day or twice a day. The doses were increased from 30 mg per day to 240 mg per day until blood pressure was adequately controlled. Blood pressure and heart rate were measured at the same time on everyday, and the diurnal variation of blood pressure was investigated on the last day of the observation period and on the day when blood pressure was adequately controlled in the treatment period. The following results were obtained: 1) The blood pressure was reduced significantly throughout the 24 hours. Once a day dosage was found to have a hypotensive effect nearly equal to that of twice a day dosage, suggesting that KB-944 is a long-acting drug. 2) The degree of daily variation of blood pressure in the treatment period was not significantly different from that in the observation period. 3) Side effects such as headache and heavyheadedness were transiently observed in 16% (6/ (37)) of the patients treated with both the once and the twice a day dosing regimens. 4) As to hypotensive effect, the rates of " remarkable fall" and "fall" were 63% (12/ (19)) for the once a day dosage and 72% (13/ (18)) for the twice a day dosage.
