I reported the new method. of Insulin assay which is determinated in vivo with hypophysectomized-Adrenalectomized rats. Insulin which is used for assay is extracted from the peripheral venous blood. <BR>1) Employed animals <BR>Rats weighing 100 ± 10 gm were used, and their hypophyses were extracted. Their both adrenal glands were extracted on 7-10 days after hypophysectomy. On 2 days after the operations mentioned above, they were used for experiment without giving them any food for 3 to 4 hours. <BR>2) Standard curve <BR>The known concentration of Insulin were injected into the pertoneal cavity of hypophysectomized-adrenalectomized rats as described above, the blood sugar was measured before the injection and one hour later. The blood sugar decreasing rate was estimated by those values and its standard curve was made. The curve shows an approximate linear relationship between the range of 0.2-3.0.mu. <BR>3) Method of extracting Insulin from their plasmas. <BR>Insulin was extracted by the method of Best Jephcott u. Scott from fresh plasma. Injecting a certain dosis of Insulin into the pertoneal cavity of hypophysectomized-adrenalectomized rats and determinating that blood sugar decreasing rate, I estimated the Insulin level of the sample from the standard curve. <BR>4) Discussion of the extraction method <BR>I didn't find any fall in blood sugar in the extracted sample which was conducted by cystein hydrochloride and alkali. 86.6% of Insulin I<SUP>131</SUP> was measured in the extract of the normal human plasma added with Insulin I<SUP>131</SUP>. <BR>5) Insulin level in the plasma of normal human and diabetic patient <BR>In 13 cases of normal people in hunger the plasma Insulin level was 1-5m.u./cc.. In diabetic patient of fatty and normal type in hunger its level was 2-0.5m.u./cc..
