The effect of eel calcitonin (ECT) on cultured mononuclear cells from giant cell tumor of bone was examined morphologically. The cells were found to possess receptors for ECT by immunohistochemical techniques, and intracellular cyclic AMP (cAMP) content increased by ECT treatment. Fresh medium containing 1.0U of ECT was changed daily, which continued for 10 or 20 days. Whether the cells were affected morphologically by ECT was examined using transmission and scanning electron microscope. Neither degenerative changes nor necrosis was seen.The effect of ECT on multinucleated giant cells and mononuclear cells seen in vivo was not produced in vitro. One of the reasons for this result seemed to be the absence of multinucleated giant cells in passaged cultures.
