An osteosarcoma cell line was established and characterized with respect to several metabolic parameters. The cells were originated from a case with osteosarcoma of the femur of a 14-year-old girl. The surgical specimen of the tumor showed histologically that the tumor comprised anaplastic cells, osteoblasts, tumorous osteoid and chondrocytes. The cultured cells revealed biologic properties of osteoblasts and preosteoblasts. Alkaline phosphatase activity was identified on the cytoplasmic membrane of the cells. Deposition of calcium around the cells and production of Type I collagen were identified. Injection of the cells into nude mice gave rise to the tumor histologically similar to the original. The cells are useful as a model for the study of mechanism of bone metabolism in vitro, as well as for the study of the treatment of osteosarcoma.
