Establishment and Characterization of the Two New Cell Lines derived from Metastatic Human Renal Cell Carcinoma.

New cell lines, termed as KMR-1 and KMR-2, were established fron metastatic bone tumor of human renal cell carcinoma (clear cell type). They have been passaged in vitro for about 8 months. Doubling times of KMR-1 and KMR-2 were respectively 32 and 51 hours. These cells have retained glycogen (stained by periodic acid Shiff reaction and destained by diastase) and fat drops (stained by oil red O stain procedure), which are characteristic to original tumor cells.Matrix metalloproteinase (MMP) -2 and MMP-3 were detected in the conditioned medium using SDS-PAGE immunoblotting technique. Two bands of MMP-2 (Molecular weight [Mr] of 72KD and about of 30KD) were detected from KMR-2 and MMP-3 (Mr of 57KD) was detected from KMR-1. These MMPs seem to relate the metastatic potentiality of renal cell carcinoma cells.
