Enviomycin (Tuberactinomycin) の非定型抗酸菌に対する抗菌力 : その1試験管内抗菌力

Enviomycin (EVM) was already established as the antituberculous drug, however, some important problems remain to be studied. One of them is antibacterial activity for atypical mycobacteria. We dont have any surely effective drug at present for atypical mycobacteria in spite of the increase in the number of atypical mycobacterial diseases. From this point of view the effect of EVM for atypical mycobacteria was studied. In this report antibacterial activity in vitro was reported.<BR>1. Materials and method<BR>Seven strains of <I>M. kansasii</I>, 6 strains of M. scrofulaceum, 16 strains of M. intracellulare, 1 strain of M fortuitum and 1 strain of M. chelonei were employed as atypical mycobacteria.<BR>0.01 mg equivalent dose of Dubos liquid culture of each strain was inoculated on each medium.<BR>Sensitivity to EVM was examined at the concentration of 5, 10 and 20 μg/m/ for Dubos liquid media and Kirchner semiliquid agar media with 10% horse serum, and at 25, 50 and 100 μg/m<I>l</I> for 1% Ogawas media.<BR>In the second experiment sensitivity to EVM, SM and KM was examined at the same time by 1% Ogawas media at the concentration of 25, 50 and 100 μg/m<I>l</I>.<BR>2. Results<BR>Sensitivity to EVM of atypical mycobacteria in Dubos media is shown in Table 1, in Kirchner semiliquid agar media in Table 2, and in 1% Ogawas egg media in Table 3. Results were observed at the third week. As shown in Table 1, in Dubos media all three strains of <I>M. kansasii</I> and 3 among 4 strains of M. scrofulaceum were sensitive to 5 μg/m<I>l</I> of EVM, but only 2 among 11 strains of M. intracellulare were sensitive to 5 μg/m<I>l</I>.<BR>As shown in Table 2, in Kirchner semiliquid agar media 2 among 3 strains of <I>M. kansasii</I>, 1 among 4 strains of M. scrofulaceum and only 1 among 11 strains of M. intracellulare were sensitive to 5μg/m<I>l</I>. Nine strains of M. intracellulare and rapid growers (<I>M. fortuitum</I> and <I>M. chelonei</I>) were resistant to 20μg/m<I>l</I>.<BR>As shown in Table 3, in 1% Ogawas media no strain of <I>M. kansasii</I> and only 1 strain of <I>M. scrofulaceum</I> were sensitive to 25 μ/ml. Growth of all strains of M. intracellulare was not inhibited at all at 25μg/m<I>l</I>, but 3 among 11 strains were sensitive to 100μg/m<I>l</I><BR>M. fortuiturn and M. chelonei were resistant to 100μg/m<I>l</I>.<BR>In the second experimet sensitivity to EVM, SM and KM of each 5 strains of <I>M. kansasii</I>, M. scrofulaceum and M. intracellulare were examined by 1% Ogawas media at the concentration of 25, 50 and 100μg/m<I>l</I> of each drug.<BR>Each strain was the isolated one from the patient who was never given each drug.<BR>As shown in Table 4, EVM was more effective generally than SM and KM for the strains of <I>M. kansasii</I> and M. scrofulaceum, however, all strains of M. intracellulare were resistant to 100μg/m<I>l</I> of each drug.<BR>3. Conclusion<BR>Antibacterial activity of EVM to atypical tnycobacteria was studied in vitro.<BR>1) Antibacterial activity of EVM depended on media. The highest activity was shown in Dubos media, next Kirchner semiliquid agar media with 10% horse serum, and activity was remarkably decreased in 1% Ogawas media.<BR>2) A small number of strains of M. intracellulare was sensitive to EVM.<BR>3) EVM was proved to be effective for strains of <I>M. kansasii</I> which were isolated from patients who have never been treated with EVM, SM and KM.
