Abnormal high level of proteasomes in sera from patients with hepatoma.

Proteasomes are new ring-shaped, large protease complexes with multicatalytic activities. We examined their levels in sera from patients with liver cirrhosis, metastatic liver carcinoma and hepatocellular carcinoma by sandwich enzyme immunoassay with their monoclonal and polyclonal antibodies. The levels of proteasomes in sera from 50 healthy adults were uniformly low, whereas the levels of proteasomes in sera from patients with metastatic liver carcinoma and hepatocellular carcinoma were high. When the neoplastic parts of metastatic liver carcinoma or hepatocellular carcinoma were removed by hepatectomy, the levels of proteasomes in sera from patients were decreased. Statistical analysis showed no significant correlation between the levels of proteasomes and α-fetoprotein or CEA. These results suggest that the measurement of the levels of proteasomes in sera might be useful for the diagnosis of hapatoma.
