The contents of saikosaponins, baicalin, cinnamic aldehyde and cinnamic acid differ among the decoctions and the extracts of the traditionally prescribed medicine, "Saikokeishi-to" The decoctions were prepared either (1) in an earthen tea pot according to the traditional procedure or (2) in a flask under reflux. The extracts were prepared by treating the materials with (3) water, (4) 20% ethanol or (5) 99% ethanol, followed by the evaporation of the solvent under reduced pressure. The quantity of the cinnamic aldehyde was 55.1% in the decoction 2, while in the decoction 1 it was 8.9% of the decoction 1, probably due to the loss by the steam distillation during the preparation. In the extract 3, only a trace of the compound was detected, as it was almost completely removed during the evaporation of water. The contents of saikosaponin and baicalin in the decoction 2 were slightly less than those in the decoction 1; they were not lost by the evaporation of water. Extraction of the materials with 20% ethanol increased markedly the quantity of the extracted saikosaponins, but increased only slightly the yields of bacialin and the total extractables. Extraction with 99% ethanol reduced both the total yield of extractables and the quantity of baicalin markedly. The contents of saikosaponin a and d in the extract 5 were apparently less than those in the extract 4, but were slightly more than those in the two types of decoctions and the extracts 3 and 5. The quantity of cinnamic aldehyde originally present, amounting 22%, was recovered in the extract 5. The addition of ethanol inhibited the conversion of saikosaponin d into b_2. The formation of cinnamic acid from cinnamic aldehyde during the extraction was also inhibited with ethanol.
