The Relationship Between Serotonin 2A Receptor Gene Polymorphism and Alzheimer's Disease

Objective: Postmortem and biopsy studies of the brains from patients with Alzheimer's disease(AD) show a decrease in the levels of serotonin (5hydroxytryptamine, 5-HT), of 5-HT receptors, and of 5-HT transporter sites. The purpose of the present study was to confirm the presence of an association between the 5-HT2A receptor gene and the etiology of AD. Methods : The present casecontrol study analyzed 5HT2A receptor polymorphism, 102T/C and -1438G/A, and apolipoprotein E (ApoE) polymorphism in 164 patients with AD and 164 age- and gender-matched healthy controls. Results : A complete linkage disequilibrium was observed between the 102 T/C and -1438 G/A polymorphisms. The increased frequency of the 102C allele in AD patients was highly significant. There was a corresponding significant difference in the genotype distribution between AD patients and controls. The creation of subgroups based on the presence or absence of the ε4 allele showed a significant association between 5HT2A receptor gene polymorphism and AD without the ε4 allele. Moreover, a slight but significant statistical association was also observed between the allele of the 5HT2A receptor gene and AD with the ε4 allele. Conclusion : It is likely that the 5HT2A receptor polymorphism is involved in the pathogenesis of AD and this polymorphism thus plays a substantial role in conferring susceptibility to AD. A synergic effect of 5HT2A receptor polymorphism and the ApoE polymorphism also probably contributes to AD susceptibility.
