Our studies on the constituents of solanaceous plants resulted in the isolation of many steroidal glycosides, such as spirostane, furostane, solasodane and solanidane glycosides, and some of which were found to exhibit anti-cancer and anti-HSV-1 activities. As a further extension of these studies, we have investigated on the constituents of Solanum cilistum and have isolated nineteen withanolide-type steroids which were different pattern from the above compounds so far obtained from Solanum plants. A major component among them, SCL-a, was characterized as 17α-hydroxy-1-oxo-24,25; 22,26-diepoxy-(20R,22R,24S,25R,26R)-ergost-2,5-diene on the basis of spectroscopic properties and X-ray crystallographic analysis and other compounds were also assigned as having closely related structures with that of SCL-a. They are classified for convenience into five categories on the basis of the biogenetic consideration: 1) Ergostane Type-SCL-v is regarding as a starting key material to be converted into withanolide compounds. 2) 1,3-Diol Type-SCL-t, -i and -j are supposed to be the precursors of 1-one-3-O-sulphate type. 3) 1-One-3-O-sulphate Type-SCL-12 and -w are conceivable to be intermediates to bear the 1-one-2-ene type and 3,5-cyclo type. 4) 1-One-2-ene Type-SCL-a, -b, -d, -q, -r, -s, -f, -g and -h were the most abundant components in this plant. 5) 3,5-Cyclo Type-SCL-p, -pm, -pl and -u are suggested to be derived from 1-one-3-O-sulphate type. They are the first finding of the withanolide-type steroids from the Solanum genera. As for the representative some specimens, we examined the screening by the panel of the man-cultured cancer cells and anti-HSV-1 activity. Their results would be discussed.
