Different expressions of growth factor mRNAs in keloids and hypertrophic scars

Objective : Keloids are regulated by various cell growth factors. Transforming growth factor beta (TGF-beta) is well known for the leading role in fibrosis. It is thought that connective tissue growth factor (CTGF) enhanced fibrosis caused by TGF-beta. The present study investigated the expressions of TGF-beta1, TGF-beta type II receptor and CTGF mRNAs in keloid and hypertrophic scar cases. Materials and Methods : Routinely 10% formalin-fixed and paraffin-embedded sections of specimens of 6 keloids, 4 hypertrophic scars, and 4 mature scars from excisions at the Kitasato University Hospital were used in this study. The expressions of TGF-beta1, TGF-beta type II receptor and CTGF mRNA were studied by the highly sensitive in situ hybridization method with digoxigenin labeled cRNAprobes. The expression of beta-2-microglobulin mRNA was used as an internal control to confirm the preservation of mRNA. Results : In generally, high levels of TGF-beta1 mRNAs were observed in the fibroblasts of keloids, but no or low levels of mRNAs were detected in those of hypertrophic and mature scars. High levels of CTGF mRNA in fibroblasts were observed both in keloids and hypertrophic scars however only low levels could be detected in mature scars. Conclusions : We observed the form of fibroblasts of keloids and hypertrophic scars. We noted a significant difference in the expression of cell growth factors mRNAs by changes in the cell forms.
