Association of Tyrosine Nitration after Intratracheal Lipopolysaccaride Administration with Myeloperoxidase Activity in Rat Lung

Previous studies have suggested that lipopolysaccharide (LPS)-induced lung inflammation results in tyrosine nitration. Tyrosine nitration is thought to occur via a peroxynitritepathway or a peroxidase-pathway. In most previous studies, however, a peroxynitrite-pathway has been assumed. The aim of this study was to evaluate whether the tyrosine nitration that occurs after the intratracheal administration of LPS is the result of a peroxynitritedependent pathway or a myeloperoxidase (MPO)-dependent pathway. To determine the mechanism of nitration, lung tissue from leukocyte-depleted rats was immunohistochemically stained for inducible nitric oxide synthase (iNOS), nitrotyrosine and chlorotyrosine (a byproduct of the MPO-dependent pathway) after the intratracheal administration of LPS. The total cell count and type of cell differentiation present in the bronchoalveolar lavage fluid (BALF) and the MPO activity in the lung homogenate were also measured. iNOS, nitrotyrosine, and chlorotyrosine staining were enhanced in the cells of the alveolar wall, alveolar macrophages and the neutrophils of LPS-treated rats, iNOS staining after LPS treatment was also enhanced in the cells of the alveolar wall and alveolar macrophages in leukocyte-depleted rats; nitrotyrosine and chlorotyrosine staining were not observed in these cells. Tyrosine nitration was associated with the neutrophil counts in the BALF and the MPO activity in the lung homogenate. These findings suggest MPO-dependent pathway contributes to the tyrosine nitration in the lungs of LPS-treated rats and that leukocyte depletion reduces tyrosine nitration as well as chlorination by inhibiting neutrophil accumulation in the lungs.
