Inhibitory Effects of Nitric Oxide (NO) on Fast Axonal Transport in Cultured Rat Hippocampal Neurons

Nitric oxide (NO) has been implicated as a mediator of neurodegeneration and neuronal cell death in the central nerves system (CNS). In the present study, we investigated the effects of NO on axonal transport in cultured rat hippocampal neurons. We used video-enhanced microscopy to observe particles moving through a neurite of the 7-day-cultured hippocampal neuron obtained from the 16-day-old fetal rat. The NO donor isosorbide dinitrate (ISDN) rapidly and significantly reduced axonal transport in anterograde and retrograde directions. The newly developed NO-releasing agent (±) -N-[(E)-4-ethyl-2-[(Z)-hydroxyimino]-5-nitro-3-hexene-1-yl]-3-pyridine carboxamide (NOR4) also inhibited axonal transport in a similar manner to ISDN. The NO synthase (NOS) inhibitor N-nitro L-arginine (L-NNA) and the NO scavenger 2-(4-Carboxyphenyl)-4,4,5,5-tetramethylimidazoline-1-oxyl-3-oxide (carboxy-PTIO) both rapidly and slightly activated anterograde and retrograde axonal transport. These results suggest that both exogenously applied NO and endogenously released NO inhibit axonal transport in cultured hippocampal neurons. The present study may provide the insight into the pathogenesis of brain ischemia, trauma, and CNS neurodegenerative diseases.
