Application of Acrylamide Embedding Method to Histological and Histochemical Observations

We devised a new embedding method using acrylamide gel. This new embedding method was efficiently applicable to the immunohistochemical studies on both levels of the light microscopy and the electron microscopy, as reported elsewhere. In this paper, we present other applications using this acrylamide embedding method. The method is useful for the rapid embedding method, since the all reagents using in the acrylamide embedding method are small molecules, less viscous and soluble in water. However, the ultrastructures obtained by the method are obscure, so that we come a conclusion that the acrylamide embedding method is inadequate for the histological studies on electron microscopic levels. But, the new method has a strong point; the embedding media can be removed under the mild conditions. This merit makes the macro-molecules penetrate rapidly into the tissue sections, so that the histochemical staining of glucose oxidase turns to be possible by adding exogenous peroxidase and capture reagents into the reaction mixtures. The merit permits the cellular localizations of sugar residues by using the labeled agglutinin. Thus, the new embedding method using acrylamide gel is useful for the "two-step" enzyme-histochemistry and the localization of sugar residue using the labeled agglutinin as well as for the immunohistochemical studies.
