Immunohistochemical Localization of Pyruvate Kinase Isozymes in Various Organs of Rat

Three electrophoretically isozymes of pyruvate kinase termed M_1, M_2 and L are detected in various organs of rat. The antibody against pyruvate kinase L does not cross-react with that of pyruvate kinase M. Using the difference in immunological property, the distribution of each isozyme in various organs was investigated by the immunohistochemical technique. In liver, pyruvate kinase M was predominantly recognized along the sinusoid, while pyruvate kinase L was present in the parenchymal cells. In kidney: Pyruvate kinase M was predominantly localized in the glomerulus and slightly in the proximal convoluted tubule. Pyruvate kinase L was present diffusely in the cytoplasm of the cells of proximal tubules. In intestine: Pyruvate kinase L was recognized in the cytoplasm of the intestinal absorptive cells, while pyruvate kinase M was present in the connective tissue of lamina propria mucosae. In spleen: Pyruvate kinase M was recognized in the cytoplasm of reticular cells, but pyruvate kinase L was not recognized in anywhere.
