Relation between Polymorphism of δ-Aminolevulinic Acid Dehydratase and Some Parameters in Lead Workers

δ-Aminolevulinic acid dehydratase (ALAD) is a heme biosynthetic enzyme whose inhibition by lead is one of the most sensitive indicators of lead exposure, but it is known that there is a considerable interindividual variation in blood lead concentrartions and in the health effects of lead among leadexposed workers with similar exposure levels. In man a genetic polymorphism for ALAD has been documented^<1)>, and this finding prompted us to investigate the potential for a genetically determined different susceptibility to lead. Previous reports have associated the variant ALAD allele with higher blood lead concentrations among relatively high lead exposures^<2)>, but the same association was not observed at low-level exposure^<3)>. In this study, we report the relation between ALAD genotype and some biological parameters, such as blood lead, erythrocyte ALAD activity and urinary δ-aminolevulinic acid (ALA-U) excretion in lead workers.
