Alterations in Protein Synthesis in vivo Chilling Sensitive Mung Bean Hypocotyls Caused by Chilling Stress

The effects of chilling on protein synthesis in vivo in etiolated hypocotyls of Vigna radiata L. were investigated. After exposure of the tissues to 0℃ for various periods of time, proteins were labeled with [^<35>S]-methionine at 26℃. The total amount of ^<35>S incorporated into soluble and membrane proteins was reversibly reduced by chilling for 24 h, during which time the tissues suffered no injury. Further prolonged chilling produced an irreversible decline both in the incorporation of radioactivity and in cell viability as assessed by the extent of leakage of electrolyte. The ^<35>S-labeled proteins in the soluble and the total membrane fractions were analyzed quan-titatively. Chilling of etiolated hypocotyls for one or two days induced the syntheses of two soluble proteins (82 and 74 kDa) and one membrane protein (80 kDa). Moreover, three heat-shock proteins (HSPs) that were induced by heat stress (41℃, 4 h) had the same electrophoretic mobilities as those of the proteins induced directly or indirectly by the chilling treatment.
