Structural changes of pea chromatin due to in vitro or in vivo treatment with Al were investigated. The absorption spectrum showed that chromatin might be condensed and/or aggregated with Al treatment. Analysis of the melting profile showed that chromatin prepared from Al-treated roots was more stable to thermal treatment. Chromatin from Al-treated roots and chromatin treated~with Al in vitro were less susceptible to DNase II digestion. When the partially digested chromatin with DNase II was chromatographed with Bio-Gel A5m, the digestibility of chromatin obtained from Al-treated roots was less than that from control roots. Furthermore, most of the Al was associated with a chromatin fraction which was minimally digested with DNase II and of large size after the gel chromatography. The results suggested that Al absorbed by pea roots is somehow related to alteration of the chromatin structure, i.e. its condensation and/or aggregation. The conclusion reached was that Al toxicity results from disturbance of the nuclear activity.
