A nitrate reductase-inactivating factor (NR-IAF) was detected in a crude extract from 8-day-old barley (Hordeum distichuln L. cv. Daisen-gold) leaves by chromatofocusing. The factor seemed to be a proteolytic enzyme with a cysteine residue at its active site because l) it was thermo-labile, and trypsin treatment caused loss of activity; 2) p-chloromercuribenzoic acid and iodoacetamide inhibited its activity; 3) leupeptin, an inhibitor of trypsin-like enzymes, also inhibited its activity; and 4) proteolytic activity toward azocasein was detected for the factor preparation. The factor did not affect the activities of nitrite reductase, glutamate dehydrogenase and xanthine oxidase.
