Physiological detection of a binding substance for the agglutinability-inducing pheromone, α substance-I in Saccharomyces cerevisiae

We found a substance which binds to a substance-I to inactivate the biological activity of the pheromone to induce sexual agglutinability of a mating type cells. Both living and boiled cells of the a mating type had the a substance-I-absorbing action. The absorbing action of living cells was detected almost equally at both 0 and 28℃. Cell extract of the a mating type showed the a substance-I-inactivating action. The biological activity of a substance-I inactivated by shaking with cell-free culture medium of the a mating type was recovered by heating at 100℃, which destroyed the inactivating action of the culture medium with little effect on the a substance-I activity, indicating that a substance in the culture medium inactivated a substance-I by binding to it. This is supported by the fact that the inactivating action completely stopped in 30 min, leaving a considerable amount of active a substance-I, when the concentration of the inactivating substance was low compared with that of a substance-I. The ability to produce the binding substance was specific to the a mating type. The binding sub-stance was different from the a agglutination substance responsible for sexual agglutination.
