A new good method for the determination of ammonia by the use of o-phenylphenol was proposed. The analytical procedure is as follows : A sample solution (0.5 ml) is pipeted into a glass-stoppered test tube, to which 0.25 ml of 1% sodium nitroprusside, 1.0 ml of 5% dimethylformamide solution of o-phenylphenol, and 1.0 ml of 1% sodium hypochlorite are added successively. The solution is mixed thoroughly, warmed at 37° for 30 min, cooled under running water, and diluted with 3.0 ml of dimethylformamide. This mixture is shaken strongly until the whole solution becomes entirely clear, and the absorbance is estimated at 690 mμ against the reference of reagent blank. Standard deviation and apparent molar extinction coefficient of this method were 2.4% and 6.72×10^4,respectively. Therefore, the present method is more advantageous than other up-to-date methods for determination of ammonia in the light of three criteria, sensitivity, reproducibility, and color stability.
