The in vitro Effects of Cyclic Nucleotides on Prostaglandin-Induced Ovulation of Goldfish (Carassius auratus)(Endocrinology)

The inhibitory effects of dibutyryl CAMP (DBcAMP), 3-isobutyl-l-methylxanthine (IBMX) and forskolin on prostaglandin F_<2α> (PGF_<2α>)-induced ovulation in the goldfish (Carassius auratus) were investigated using an in vitro incubation system. Oocytes that had undergone germinal vesicle breakdown and follicular detachment in vivo, were incubated in the presence of 10.0, or 1.0 μg/ml PGF_<2α> and IBMX, DBcAMP or forskolin. PGF_<2α> alone stimulated 82% and 70% ovulation at 10.0 and 1.0 //g/ml respectively. At 10 μg/ml PGF_<2α>, 5.0mM DBcAMP, 1.0mM IBMX and 1.0 or 0.1 μM forskolin significantly blocked ovulation. At 1.0μg/ml PGF_<2α>, 5.0 or 0.5mM DBcAMP, 1.0 or 0.1 mM IBMX and 1.0, 0.1 or 0.01 μM forskolin significantly blocked ovulation. The results indicate that some point in the process of goldfish ovulation after follicular detachment, is inhibited by CAMP.
