Interaction of F-actin, Spectrin and α-Actinin

Interaction of F-actin, spectrin and α-actinin was investigated using techniques of flow birefringence and electron microscopy. Spectrin bound to F-actin resulted in an increase in the amount of flow birefringence. Mg ions, not Ca ions, were required for the binding of spectrin of F-actin. The spectrin-bound F-actin became less easily depolymerizable than intact F-actin, when ionic strength was lowered. Electron microscopic observations revealed that spectrin caused formation of random nets of F-actin and abolished the formation of F-actin bundles by α-actinin. A possible role of the interaction of these three proteins in cell motility is discussed.
