Oocyte maturation in starfishes is known to be induced by 1-methyladenine (1-MeAde) produced in the follicle cells under the influence of a gonad-stimulating hormonal peptide of neural origin (see Kanatani, 1973). In the present study, the effect of pretreatment of isolated starfish oocytes with calciumfree seawater (CaFSW) on 1-MeAde-induced oocyte maturation was investigated. Isolated oocyte of Asterina pectinifera obtained by tearing ovarian fragments with fine forceps were transferred to CaFSW and their follicles were removed by repeated pipetting. Some of these oocytes were exposed to CaFSW for 15 minutes during the above treatment, and transferred to seawater containing 1-MeAde at various concentrations from 1.6×10^<-9>M to 2×10^<-7>M and used as control oocytes. The rest of the oocytes were kept in CaFSW for 2 hours, then transferred to seawater containing 1-MeAde, and used as experimental oocytes. The degree of germinal vesicle breakdown was observed after 1 hour. As shown in Table 1, pretreatment with CaFSW for 2 hours enhanced the sensitivity of Asterina oocytes to 1-MeAde. For determining the amount of 1-MeAde contained in a given tissue or organ, bioassay of tissue extract with isolated starfish oocytes, using authentic 1-MeAde as a standard reference, is excellent since the method is very sensitive. In this respect pretreatment of isolated oocytes with CaFSW is recommended for determining a very minute amount of 1-MeAde since it enhances oocyte sinsitivity to the substance.
