Starfish oocytes within the ovary remain immature, while spermatocytes undergo spermatogenesis long before their release. According to Heilbrunn et al. (1951, 1954), testis as well as ovary contains an antimitotic substance which inhibits cell division of Chaetopterus and Arbacia eggs and seems to block meiosis in the starfish. The present study was conducted in order to examine the effect of sea water extract of ovary and testis of Asterias amurensis on the maturation of isolated oocytes. In this species, oocytes isolated in sea water usually undergo spontaneous maturation without any additional treatment. To prepare the ovary extract, lyophilized ovaries from five females were separately homogenized in a small amount of cold sea water (40mg/ml) and centrifuged at 30, 000×g for 50 minutes (3℃). The supernatants were serially diluted twice with sea water (up to 1mg/ml) and assayed with isolated oocytes. Samples of testis extract were prepared and assayed in the same way. The oocytes were isolated by tearing an ovarian fragment with forceps after treating it with calcium-free sea water for 5 minutes. They were transferred into normal sea water within 2 minutes and then kept in small Petri dishes containing the test solutions (19℃). The number of immature oocytes still having intact germinal vesicles was observed after one hour. Oocytes kept in calcium-free sea water throughout and those transferred into normal sea water served as controls. From the results shown in Table 1 it is concluded fhat sea water extract of starfish ovary contains some substance which blocks oocyte maturation; the rate of germinal vesicle breakdown decreased as the concentration of extract rose, thus confirming the result of Heilbrunn et al. (1954) with respect to the ovary extract. However, testis extract showed no such activity at the concentrations examined. These results seem to support the notion that gametogenesis is inhibited in the starfish gonad by the presence of such a substance.
