Embryos of the ascidian, Halocynthia roretzi, at early cleavage stages up to the 64-cell stage were continuously treated with cytochalasin B, colchicine, or colcemid, and cleavage of the embryos was arrested and they did not hatch out. However, early gastrulae(7.5 hr of development) and embryos at later stages that had been permanently arrested with the inhibitors were found to be able to hatch out at almost the same time (about 30 hr after fertilization)as normal embryos. This finding is additional evidence for differentiation with-out cleavage. The significance of this finding for further studies on the mecha-nism involved in the hatching of ascidian embryos is discussed.
