Expression of CETP on human normal spleen, lymph node and malignant lymphoma

Cholesteryl ester transfer protein (CETP) is a plasma glycoprotein that facilitates the transfer of cholesteryl ester, phospholipids and triglycerides between the lipoprotein, and regulates plasma high-density lipoprotein levels. In human tissue, an expression of CETP mRNA was present in liver, spleen, adrenal gland, intestine, heart and skeletal muscle. However the physiological significance of some of these observations is uncertain. In the spleen, the major source of mRNA appears to be lymphocyte. However, the histological localization of CETP in human spleen has not been reported. In this study, we examined the expression of CETP mRNA in normal spleen, tonsil, reactive hyperplasia and Non Hodgkin Lymphoma using immunohistochemistry (IH) of CETP monoclonal antibody (mAb), in situ hybridization (ISH) and western blot and Northern blot analysis. Also, serial sections and br double stain of CETP ISH and IH findings of lymphocyte cluster differentiation markers were also examined. Immunohistochemical staining for anti CETP mAb was detected in the germinal center (GC) of normal tonsil. CETP expression area cells were positive for pan-B (CD20, not shown; CD79) but other markers (CD3, not shown; CD5, not shown; CD38) were consistently negative by serial sections and double staining. In the normal lymph node sections, strong expression in GC and weak positive cells was seen in the outerside (interfolicular zone) of mantle zone. The CETP reactivity in the case of Non Hodgkin lymphoma showed a strongly expression in the neoplastic germinal center of Follicular Lymphoma. In Diffuse Large B-cell lymphoma case, the expression showed a variable pattern, almost all diffuse with slight to moderate staining. The incidence of CETP positivity in marginal cell lymphoma cases was similar to DLBL, however in Mantle Cell Lymphoma cases was, CETP staining was lacking or very low. The expression pattern of CETP mRNA by In situ hybridization method of normal lymph tissue showed expression as which the pattern by immunohistochemistry method of CETP mAb showed the same. CETP expression was mainly GC, these result was suggested that CETP expression are only to a specific stage differentiation B lymphocyte of spleen, but not plasma cell. Expression of CETP in germinal center was suggested that appears to target the maturation process of B cells.
