Zinc Content and Expression of Metallothionein mRNA in ZnSO4-Exposed Rat Liver and Kidney : A Comparative Study with a Cadmium-Administered Group

Zinc (Zn)-induced changes in liver and kidney MT levels were studied and compared with previously obtained results from a Cadmium (Cd) administered group. Male Wistar rats were given 30 mg ZnSO4 orally every second day, fora total of either 1,13or26 doses. In each group, the relations between Zn accumulation and both metallothionein (MT) mRNA and protein level were studied by Northern blot and immunohistochemical analysis. In the Cd-administered group, the level of Cd in the liver and kidney increased, as did MT-mRNA induction. In the Zn-administered group, Zn accuumu-lation did not occur, though Zn was repeatedly administered; however, MT-I and MT-Il mRNA levels increased, compared to the control group. In the 1-doseZn group, the Zn concentrations in the liver and kidney increased slightly, a possible cause for disturbance of homeostasis. Moreover, induction of MT-mRNA expression was more pronounced than in the 13 or 26-dose groups. The correlation between Zn and MT-mRNA synthesis was stronger than with Cd, and MT-I mRNA levels exceeded MT-Il mRNA levels in Zn-induced MT expression. In the Cd-administered group, Zn levels increased in both the liverand kidney. This suggests that MT was induced by CdCl2. However, in the Zn-administered group, Zn concentration did not change though MT synthesis wasmarkedly induced, suggesting that a high level of proteins other than MT is involved in Zn transport. this protein clearly plays a role in homeostasis and absorption and excretion of Zn. Immunohistological analyses found strong MT staining in the Cdadministe-red group. Evidence of MT-mRNA and MT expression in immunostaining confirmedthis finding. However, no MT immunostaining was observed in the Zn-administered group.
