HISTOCHEMICAL STUDIES ON THE SPECIFIC GRANULES IN THE ATYPICAL SPERM SERIES OF THE POND SNAIL

Specific granules of the atypical spermatids in the pond snail were tested histochemically. There are large and small atypical sperm lines, both having these specific granules. These granules are morphologically similar and have been thought to be identical. Present study, however, reveals that they are different from each other in properties. They were equally positive to PAS and some general procedures for proteins. But, tryptophan was found only in the granules of the spermatids of small line, while tyrosine and potassium were observed in those of the large line. Definite acidity of these granules is not probable because of the negative reaction to colloidal iron. The granules of large line were stained with pyronin, Janus green B and alcian blue, but not with toluidine blue, methylene blue and neutral red. This suggests some physical relation of the dyes with the granules, rather than chemical reaction of the dyes. Complete lack of basophilia of the granules of small line may be attributed to some structural difference.
