Streptococcus mutans strain AHT (serotype g) secreted a 1, 3-α-D-glucan synthase. The enzyme with pI5.1 was highly purified from the ammonium sulfate fraction by DEAE-cellulose choromatography (two times), Bio-Gel A-5m, Octyl-Sepharose CL-4B, Mono Q (FPLC), TSK-gel G3000SW (FPLC) and TSK-gel phenyl 5PW (FPLC). The enzyme preparation gave a single protein band on analysis by PAGE, SDS-PAGE and IEF. The molecular weight was estimated to be about 150, 000 by ultracentrifugal analysis. The pH optimum of the enzyme was 6.2. The apparent Km value for sucrose and dextran T10 were 4.17 mM and 0.8 mM-glucos eqeuivalent, respectively. The enzyme synthesized water-insoluble glucan from sucrose in the presence of dextran T10, but did not synthesize water-insoluble glucan without dextran T10. The data strongly suggested that the 1, 3-α-D-glucan synthase did not synthesize any glucan in the absence of primer dextran.
