Insolubilization mechanism of water-soluble glucan was studied. 1, 3-α-D-glucan synthase solution containing primer dextran (DT 2000) and sucrose was incubated at 37℃. Water-insoluble glucan was synthesized by 1, 3-α-D-glucan synthase. At the same time, water-soluble glucan, mainly derived from DT 2000, decreased with time. Linkage analysis of glucan of this reaction mixture was made by periodate oxidation and Smith degradation. Periodate oxidation of glucan showed that 1, 3-α-D-glucoside linkages increased with time. High molecular weight primer dextran was insolubilized by 1, 3-α-D-glucan synthase more completely than low molecular weight dextran. The data strongly suggest a water-insoluble glucan having linear 1, 6-α-linkages, with random 1, 3-α-branches.
