830 Expression of Lipase Gene from Pseudomonas sp. 109 in Cell-Free System

A lipase (LipL) from Pseudomonas sp. 109 can catalyze synthesis of macrocyclic lactones in anhydrous organic solvents. A modulator gene (limL) located immediately downstream from lipL had been found essential for production of functional lipase. In cell-free system lipase with catalytic activity was synthesized using E. coli S30 extract from signal-deleted lipL gene (rlipL) in the presence of its modulator that was purified from the over expressing E. coli cells. The specific activity of the in vitro expressed lipase was higher than that of refolded lipase which was first produced in E. coli as inclusion body and then reactivated by in vitro solubilization and renaturation. Active lipase could also be produced in vitro by co-expression of the lipase gene (rlipL) and the modulator gene (rlimL).
